BACKGROUND: Staphylococcus aureus is a cosmopolitan and pathogenic microorganism associated with various diseases spectra and antimicrobial resistance of public health importance. AIM: This study determined the phenotypic characteristics of S. aureus isolated from patients in healthcare institutions in Zaria metropolis. STUDY DESIGN: A cross-sectional hospital-based study was carried out in 5 healthcare institutions. Four hundred and twenty clinical samples were collected and analyzed. RESULTS: Majority of the patients (54.3%) were within the age range 21-40 years and mean age of 26.04 ± 12 years. Approximately, 70% of the respondents had history of antibiotic use prior to consultation in the hospitals and were self-prescribed, and 91.2% were outpatients. The most commonly abused antibiotics were ampicillin-cloxacillin (19.5%) and cotrimoxazole (10.0%), and the mean duration of their use was 3.5 ± 1.3 days. The detection rate for S. aureus was 10% and 5.2% for MRSA. The S. aureus isolates showed the highest frequency of resistance against ampicillin 42 (100%), followed by penicillin G 39 (92.9%) and least was to gentamicin 5 (11.9%). The frequency of resistance for the MRSA were ampicillin 22 (100%), penicillin G 21(95.5%) and least was to gentamicin 2 (9.1%). The minimum inhibitory concentrations of oxacillin were greater than 128 µg /ml. CONCLUSION: The detection rate of S. aureus and MRSA strains are of great public health concern which requires continuous health education on rational use of antibiotics among others.


CONTEXTE: Staphylococcus aureus est un micro-organisme cosmopolite et pathogène associé à divers spectres de maladies et à une résistance aux antimicrobiens d'importance pour la santé publique. OBJECTIF: Cette étude a permis de déterminer les caractéristiques phénotypiques de S. aureus isolé chez des patients dans des établissements de santé de la métropole de Zaria. PLAN DE L'ÉTUDE: Une étude transversale en milieu hospitalier a été menée dans 5 établissements de santé. Quatre cent vingt échantillons cliniques ont été recueillis et analysés. RÉSULTATS: La majorité des patients (54,3 %) étaient âgés de 21 à 40 ans et l'âge moyen était de 26,04 ± 12 ans. Environ 70 % des répondants avaient des antécédents d'utilisation d'antibiotiques avant la consultation dans les hôpitaux et étaient auto-prescrits, et 91,2 % étaient des patients externes. Les antibiotiques les plus fréquemment utilisés étaient l'ampicillinecloxacilline (19,5 %) et le cotrimoxazole (10,0 %), et la durée moyenne de leur utilisation était de 3,5 ± 1,3 jours. Le taux de détection de S. aureus était de 10 % et de 5,2 % pour le SARM. Les isolats de S. aureus ont montré la plus grande fréquence de résistance à l'ampicilline 42 (100%), suivie de la pénicilline G 39 (92,9%) et la plus faible à la gentamicine 5 (11,9%). La fréquence de résistance pour le SARM était de 22 (100%) pour l'ampicilline, 21 (95,5%) pour la pénicilline G et 2 (9,1%) pour la gentamicine. Les concentrations minimales inhibitrices de l'oxacilline étaient supérieures à 128 µg /ml. CONCLUSION: Le taux de détection des souches de S. aureus et de SARM est un grand problème de santé publique qui nécessite une éducation sanitaire continue sur l'utilisation rationnelle des antibiotiques entre autres. Bone marrow is a source of osteoprogenitor cells which are the most important factor of bone formation and healing of fracture. The aim of the study is to evaluate the outcome of bone marrow injection in the management of delayed union and non-union. This prospective study was performed in the department of Orthopaedics, Mymensingh Medical College Hospital, Mymensingh, Bangladesh from January 2017 to June 2019. In this study 21 patients with delayed union and non-union were treated by bone marrow injection. Bone marrow were aspirated from the anterior or posterior iliac crests then injected percutaneously into the fracture site. Full union was achieved in 15 cases, while failed in the others. No major complications were seen during or after the procedure. It is a safe, easy and a minimally invasive procedure compared to usual open bone graft especially for cases with high risk of anesthesia or risk of infection.

Samples for sediment and two species of native mangrove plants were collected from seven sampling sites for assessing the level of metal contamination. Results of the studied metals displayed the order of pollution as Fe > Ti > Zr > Rb > Zn > Sr > Pb > Y > Cu > Cr > As accordingly. Geoaccumulation index and contamination factor revealed that the sediment samples were unpolluted to moderately polluted by Zn, Fe, Ti, Rb, Y, and Zr. Ecological risk factor depicted a pollution-free condition in the study areas. PCA, CA, and correlation coefficient indicated that the source of the metals in the environment was anthropogenic. Bioconcentration factor values were found to be below 1 in both plant species. Conversely, transfer factor values for most heavy metals were found to be >1 in both plant species, which reflects the phytoremediation ability of plants.

RESUMO

We came across an 81 years old male with symptomatic severe aortic stenosis. He was hypertensive and had history of CABG 9 years back. Due to his advanced age and co morbidities, he was at high surgical risk. He underwent transcatheter aortic valve implantation in our centre (United Hospital Ltd) in July 2017 and no complications occurred during or in the peri-procedural period. He had good functional and haemodynamic results at 3 months follow-up.

Contaminants in road dusts can directly pose significant human health risks through oral ingestion, particle inhalation, and dermal contact. Therefore, this study has been designed to analyze heavy metal contaminations in 88 street dusts collected from the 22 high traffic sites and industrial areas of the Southeast Asian Megacity: Dhaka (capital of Bangladesh) using energy dispersive X-ray fluorescence (EDXRF) spectroscopy. This study revealed that the maximum Pb, Cd, Zn, Cr, Ni, As, Mn and Cu contents in the street dust samples were 18.9, 11.6, 239.2, 144.3, 37.1, 8.1, 261.5 and 49.6 mg/ kg respectively in which Ni and Pb concentration were 2 times, and Cd concentration was 200 times higher than the background level in soil; and As concentration was slightly higher than soil background level. The spatial distribution of heavy metal concentrations in street dust samples in Dhaka City was revealed that the hot spot areas of Pb, Ni, Cd and As were mainly associated with heavy traffic and industrial activities. The risk assessment strategies were used for this study for identifying the routes of exposure through oral ingestion, inhalation, and dermal contact by the fine particles (~75 µm) of street dust, especially for children based on the US EPA health risk models. Both non-carcinogenic and carcinogenic risks of heavy metals were characterized in street dust samples. Results based on the hazard index (HI), in the case of non-cancer effect, the ingestion of dust particles of children and adults in Dhaka City appeared to be the route of exposure to street dust that results in a higher risk for heavy metals, followed by dermal contact. This study revealed that the inhalation of re-suspended particles through mouth and nose were almost negligible. It was also noticed that children were experiencing the potential health risk due to HI for Cr (1.04), which was slightly higher than the safe level 1, and Cd (0.69) was close to the safe level 1. Reversely, cancer risk for Cr (i.e. 4.27 × 10-6) was fallen within the range of threshold values (10-4 to 10-6) and As (i.e. 9.59 × 10-7) was close to the upper limit of threshold values (10-4 to 10-6).

Nigeria, being a tropical nation, characterized by favorable climatic conditions, may display high chances of feed contaminations due to aflatoxigenic Aspergillus flavus with the consequences of health risks associated with the consumption of dairy products. A cross-sectional study was conducted to examine the risks of occurrence and distribution of aflatoxigenic Aspergillus flavus (A. flavus) and aflatoxin B1 (AFB1) contamination levels based on the European Union (EU) and United States Food and Drug Administration (USFDA) set limits. Feeds (n = 144) were collected from selected conventional and traditional dairy herds; prepared and analyzed using immuno-affinity column (IAC) and high performance liquid chromatography (HPLC) respectively. Forty eight (55.8%) isolates were identified as A. flavus of the isolated Aspergilli (n = 86). Of this proportion, 12 (25.0%) were aflatoxigenic strains. An outrageous number of the tested feeds (86.8%, n = 144) were positive for AFB1 contamination. Detectable AFB1 concentrations ranged between (0.5 and 24.8) µgKg-1 and were distributed variously according to many factors of distribution. Eighty three (66.4%, n = 125) of the AFB1 contaminated feed samples showed AFB1 concentrations between 5 and ≥20 µgKg-1. All-inclusive policies are key to reducing the health risks posed to the consumers of dairy products.

A cross sectional study was conducted to determine the seroprevalence and risk factors of brucellosis and Q fever in cattle in Maigana and Birnin Gwari agro-ecological zone of Kaduna State, Nigeria. This study aimed at determining the significance of Brucella spp. and Coxiella burnetti infections in cattle. A total of 400 sera samples (139 from males and 261 from females cattle) were collected and screened for brucellosis using Rose Bengal Plate test (RBPT) and competitive enzyme linked immunosorbent assay (cELISA) for brucellosis and indirect enzyme-linked immunosorbent assay (iELISA) for Q fever. A structured questionnaire was used to collect data on the sampled animals from the study population. Data were analyzed to determine association and risk factors. Sera analysis revealed that, 18.5 and 6.8% were seropositive by RBPT and cELISA for brucellosis, while 6.2% was seropositive by iELISA for Q fever. A significant association was detected between cattle sex and sensitivity of RBPT for detecting Brucella. Meanwhile, a non-significant association was found between cattle age and breed with sensitivity of RBPT, cELISA, and iELISA. The study indicates that brucellosis and Q ever exist with high prevalence particularly among female cattle. This presents a serious public health problem, calling for greater awareness among stakeholders and for co-ordinated surveillance for the diseases among cattle populations in Nigeria.

It is considered that cow's milk is almost complete food for human as it provides most of the micronutrients and macronutrients. The cow's milks are essential for the growth and development especially for children. The main compositions of cow's milk are protein, fat, carbohydrates, vitamins and minerals which are well defined. Presently, the study of micronutrients and toxic elements in cow's milk has been widely carried out particularly in the industrialized and polluted regions because of its possibility of contamination, and thereby health risk of the consumers. The elemental composition in local cow's milk samples in Bangladesh is not well studied yet. The present study was therefore aimed to determine the level of heavy metals (Cr, Ni, As, Cd, Hg, Pb, Mn, Cu, Zn and Fe) in cow's milk using EDXRF technique. Subsequently, the experimental data was used to calculate the human health risk through the intake of both powder and liquid cows' milk available in Bangladesh. The results showed that powder milk contains significantly higher concentration of heavy metals than liquid milk samples. The HRI (health risk index) and HI (hazard index) values for most of the elements in all milk samples were within the safe limit (<1.0) or close to safe limit (≤1.0) with an exception of Hg. However, HRI value for Hg in powder milk samples for both children and adult showed a value higher than one (>1). MPI (metal pollution index) value for powder milk samples are very high compared to other type of milk samples analyzed in this study. Therefore, it has been suggested that heavy metal contamination through local powder milk samples might have significant negative impact (threat) on human health.

An assessment of the dietary risk of heavy metal exposure to humans is important since it is the main source of exposure. This study aimed to estimate the degree of contamination and assess the probable health risk in the prawn food chain. In prawn feed, the concentrations of metals were detected in the following order: Hg > Co > Pb > Cd. The concentrations of heavy metals in prawn were the highest for Co and lowest for Cd. Trace amounts of As and Cr were detected in the analyzed sample. Target hazard quotients for heavy metals for adults were >1 for Pb, Cd, Hg, and Co, and for children, the same were high for Co and Hg, indicating significant health risks upon dietary exposure. All the prawn samples contained nine-fold and fourteen-fold higher concentrations than the maximum acceptable levels for Pb and Hg, respectively (0.5 mg kg-1; WHO/FAO). Human health risk due to the Co exposure is quite alarming as the level of exposure was found to be very high. In the prawn samples intended for human consumption, the hazard index (HI) was highest in the samples obtained from Bagerhat (3.25 in flesh and 3.26 in skin), followed by the samples obtained from Satkhira (2.84 in flesh and 3.10 in skin) and Dhaka City Corporation (2.81 in flesh and 3.42 in Skin); this indicates a potential risk of prawn consumption obtained from Southeast Bangladesh. This is particularly problematic as this area accounts for the majority of prawn production and export of the country.

In this study, Listeria (L.) monocytogenes isolated from milk and milk products in Kaduna, Nigeria, were subjected to a multiplex PCR assay to identify virulence-associated genes (such as prf A, inl A, hly A, act A, and iap). Of the 36 isolates, 9 (25%) were positive for one or two virulence-associated genes. Based on the sample type, 6 (16.9%) of the isolates that possessed virulence-associated genes were obtained from raw milk, 2 (3.2%) from "Manshanu," and 1 (2.8%) from "Kindrimo." Sequence and phylogenetic analysis based on the 16S rRNA revealed that Nigerian L. monocytogenes isolates (NGA 34A, NGA 35A, NGA 41A, and NGA 38A), when compared with reference L. monocytogenes, were grouped into two distinct clusters, A and B, with sequence (NGA 34A, NGA 35A, and NGA 41A) phylogenetically closer to J1776; N1-011A; R2-502; J1816; and J2-031, whereas L. monocytogenes isolate (NGA 38A) clustered with EDG; J1-220; J1926; J1817; and J2-1091. The separation of the Nigerian L. monocytogenes isolates into linage A (responsible for epidemic listeriosis) and lineage B (responsible for sporadic cases of listeriosis) is of public health concern and that local isolates might have potentials for human food borne listeriosis based on the virulence factors so far identified.

The study investigated socio-cultural characteristics of pastoralists that influenced on the tsetse-trypanosome-cattle reservoir interface thereby predisposing them to HAT in Niger State, North-central Nigeria. It was a cross-sectional survey of adult pastoral herders, aged 30 years and above, and conducted between October 2012 and February 2013. A face-to-face structured questionnaire was administered on the pastoralists nested in 96 cattle herds with questions focused on pastoralists' socio-cultural activities and behavioral practices related to HAT risk. Descriptive and analytic statistics were used to describe the obtained data. A total of 384 pastoralists participated, with mean age of 49.6  ± 10.76 SD years. Male respondents constituted 86.7% of gender, while pastoralists of age group 40-49 years constituted 35.4% of respondents. About 59.4% of the pastoralists had knowledge about HAT and its symptoms and only 33.9% of them believed that cattle served as reservoir of HAT trypanosome. Knowledge/belief levels of the pastoralists about African trypanosomiasis occurrence in humans and animals were statistically significant. Males were four times more likely to be exposed to HAT (OR = 3.67; 95% CI: 1.42, 9.52); age group 60-69 was also four times more likely to be exposed (OR = 3.59; 95% CI: 1.56, 8.28); and nomadic pastoralists were two times more likely to be exposed to HAT (OR = 2.07; 95% CI: 1.37, 3.14). All cultural practices significantly influenced exposure to HAT with extensive husbandry system three times more likely to predisposed pastoralists to HAT (OR = 3.21; 95% CI: 1.65, 6.24). Socio-cultural characteristics of pastoralists influenced exposure to HAT risk and, therefore, there is a need to sensitize them to bring changes to their socio-cultural practices and perceptions to achieve effective and long term sustainable HAT control. Elimination strategies of parasites in animals and vectors should be considered to avoid reintroduction from animal reservoirs.

The use of antibiotics in poultry can result in residues in eggs. The joint FAO/WHO committee recommended banning the use of chloramphenicol (CAP) in food animals due to its public health hazards of aplastic anaemia, leukaemia, allergy, antibacterial resistance and carcinogenicity. This paper determines the prevalence of CAP residues in chicken eggs and assesses the usage and awareness of its ban amongst poultry farmers in the Federal Capital Territory (FCT), Abuja, Nigeria. A cross-sectional survey of registered poultry farmers in FCT was conducted using questionnaires to determine CAP administration in poultry and awareness of its ban. Pooled egg samples were collected from each poultry farm surveyed and from randomly sampled government-owned markets in FCT. Source of eggs by state were identified by the marketer at the time of collection. Samples were analysed using an enzyme-linked immunosorbent assay (ELISA) technique for the presence of CAP, and prevalence was determined. Of 288 total pooled samples collected, 257 (89.2%) were from the markets and 31 (10.8%) were from poultry farms. A total of 20 (7%) pooled egg samples tested CAP-positive; market eggs originated from 15 (41%) states of the country. Of the market eggs, 16 (6.2%) pooled samples tested positive. Of eggs from poultry farms, four (12.9%) tested positive. Mean CAP concentrations in the positive samples ranged from 0.49 to 1.17 µg kg(-1) (parts per billion). CAP use amongst poultry farmers in FCT was 75.5%; awareness of the CAP ban was 26.3%. Though 66% of veterinarians were unaware of a CAP ban, they were more likely to be aware than other poultry farmers (odds ratio (OR) = 1.4). Farm managers who use CAP were more likely to be aware of CAP ban than the farm managers not using CAP (OR = 5.5; p = 0.04). Establishing a drug residue surveillance and control program and enforcement of CAP legislation/regulation is needful to educate and prohibit the widespread CAP use amongst Nigerian poultry farmers.

UNLABELLED: The bacterial genera Listeria and Staphylococcus have been frequently isolated from food products and are responsible for a number of animal and human diseases. The aim of the study was to simultaneously isolate and characterize L. monocytogenes and Staphylococcus species from 300 samples of raw meat and meat products, to determine the susceptibility of the organisms to commonly used antimicrobial agents and to determine the presence of haemolysin A (hyl) virulence gene in L. monocytogenes and staphylococcal cassette chromosome mecA (SCCmec) gene in the Staph. aureus isolates using PCR. Of the 85 Listeria isolates tested, 12 L. monocytogenes were identified and tested for their sensitivity to 14 antimicrobial agents. All the 12 isolates (100%) were resistant to nine antimicrobial agents, but however sensitive to gentamicin. Only one isolate was found to harbour the hylA gene. Twenty-nine isolates were confirmed as Staph. aureus by the Microbact 12S identification system and were all presumptively identified as methicillin-resistant Staph. aureus species using oxacillin-resistant Staph. aureus basal medium (ORSAB). The 29 Staph. aureus isolates were tested for their sensitivity to 16 antimicrobial agents, and 11 were resistant to methicillin. None of the 11 Staph. aureus isolates harboured the methicillin resistance, mecA gene. SIGNIFICANCE AND IMPACT OF THE STUDY: Listeria monocytogenes and Staphylococcus aureus are important agents of foodborne diseases. Occurrence of these infectious agents was established in meat and meat products in Zaria, Nigeria. Majority of isolates obtained from this study, displayed multidrug resistance to commonly used antimicrobial agents, including methicillin resistance among the Staph. aureus isolates. The potential virulence of L. monocytogenes found in ready-to-eat food was documented by the carriage of hly A gene by one of the isolates. A different mechanism of methicillin resistance or different homologue of mec A gene may be circulating among Nigerian isolates.

Brucellosis and campylobacteriosis are economically important diseases affecting bovine reproductive efficiency in Nigeria. A questionnaire-based survey was conducted in 271 cattle herds in Adamawa, Kaduna and Kano states of northern Nigeria using multistage cluster sampling. Serum from 4745 mature animals was tested for Brucella antibodies using the Rose-Bengal plate test and positives were confirmed in series-testing protocol using competitive enzyme-linked immunosorbent assay. Preputial scrapings from 602 bulls were tested using culture and identification for Campylobacter fetus. For each disease, a herd was classified as positive if one or more animals tested positive. For each herd, information on potential managemental and environmental risk factors was collected through a questionnaire administered during an interview with the manager, owner or herdsman. Multiple logistic regression models were used to model the odds of herd infection for each disease. A zero-inflated Poisson model was used to model the count of Brucella-positive animals within herds, with the number tested as an exposure variable. The presence of small ruminants (sheep and/or goats) on the same farm, and buying-in of >3 new animals in the previous year or failure to practice quarantine were associated with increased odds of herd-level campylobacteriosis and brucellosis, as well as increased within-herd counts of Brucella-positive animals. In addition, high rainfall, initial acquisition of animals from markets, practice of gynaecological examination and failure to practice herd prophylactic measures were positively associated with the odds of C. fetus infection in the herd. Herd size of >15, pastoral management system and presence of handling facility on the farm were associated with increased odds, and gynaecological examination with reduced odds of herd-level Brucella seropositivity. Furthermore, the zero-inflated Poisson model showed that borrowing or sharing of bulls was associated with higher counts, and provision of mineral supplement with lower counts of Brucella-positive cattle within herds. Identification of risk factors for bovine campylobacteriosis and brucellosis can help to identify appropriate control measures, and the use of zero-inflated count model can provide more specific information on these risk factors.

BACKGROUND: Heavy alcohol consumption is a risk factor for developing atrial fibrillation, but whether chronic alcohol use affects left atrial volume is unknown. We evaluated the association of self-reported alcohol consumption with 5-year change in left atrial volume among patients with coronary heart disease (CHD). METHODS: We studied 601 participants with stable CHD who underwent 2-dimensional echocardiography at baseline (2000-2002) and after 5 years of follow-up (2005-2007). Alcohol consumption was determined at baseline with the use of the Alcohol Use Disorders Identification Test consumption questions (AUDIT-C), with a standard cutoff point of ≥3 used to define at-risk drinking. We used logistic regression to evaluate the association of baseline alcohol use with 5-year increase in left atrial end-systolic volume index (defined as being in the highest tertile of percent change). RESULTS: After adjustment for covariates, each standard deviation (2.4-point) increase in AUDIT-C score was associated with a 24% greater odds of experiencing a 5-year increase in left atrial volume index (adjusted odds ratio [OR] 1.24, 95% confidence interval [CI] 1.04-1.48; P = .02). Compared with the 369 participants who had AUDIT-C scores of <3, the 171 participants with scores of 3-5 had a 51% greater odds (OR 1.51, 95% CI, 1.11-2.25) and the 61 participants with scores of >5 a 98% greater odds (OR 1.98, 95% CI, 1.10-3.56) of experiencing a 5-year increase in left atrial volume index. CONCLUSIONS: In patients with CHD, heavier alcohol consumption is associated with a 5-year increase in left atrial volume. Whether greater left atrial volume contributes to the increased risk of atrial fibrillation associated with alcohol use deserves further study.

To evaluate the competency of Objective Structured Practical Examination (OSPE) as an assessment technique compared to Traditional Practical Examination (TDPE) in assessment of laboratory component of physiology, the results of Physiology practical examination of 400 students from 4 Medical Colleges, two Government and two non Government (Dhaka Medical College, Mymensingh Medical College, Bangladesh Medical College, Uttara Women's Medical College) under Dhaka University were studied. Students' performance in OSPE and TDPE was compared. Mean score obtained in OSPE was 77.72+/-0.66 and found significantly higher than that for TDPE (64.44+/-0.61). Again mean scores achieved in OSPE were compared among different Medical Colleges and significant difference was noted. In OSPE, male students achieved significantly higher score than that of female students, especially in responding question station. The outcome of the present study thus indicates that OSPE is a better choice as an assessment technique over the Traditional method measuring wide range of practical skill. It may be concluded that it is important for competency based performance discrimination and it also helps improving students performance quality in laboratory exercise.

OBJECTIVES: To examine how pricing policies were contrived in general dental practice in terms of fee-for-item and hourly rate and how these were affected by specialist status and the level of private care provided in a practice. DESIGN: A postal questionnaire. SUBJECTS: Members of the British Society for General Dental Surgery working in dental practice. RESULTS: Out of 160 eligible members, responses were received from 124 members (78%). Fifty-seven respondents claimed to specialise in one or more fields of dentistry. The majority of respondents consulted fellow colleagues or partners for advice on fee setting. A minority took external advice. The charging method varied according to the item of treatment with fee-for-item used predominantly for items such as a new patient examination, and hourly rate used more for items such as a direct composite restoration. Seventy-one respondents stated that their practice was 80-100% private treatment and these practitioners were significantly more likely to charge by hourly rate than fee-for-item for many items of treatment. Specialist status did not have any effect on charging method. The most important factors related to the setting of fees-for-item or hourly rate were clinical time spent, practice overheads and laboratory costs. CONCLUSIONS: This project has taken the views of a large group of experienced general dental practitioners, many of whom work purely in the private sector. The most important factors affecting fee setting were clinical time, practice overheads and laboratory costs. The method of charging was most affected by the proportion of private treatment provided by the practice.

The occurrence of antimicrobial drug residues in 500 routinely slaughtered cattle in Zaria, Nigeria, was determined. A microbial inhibition test with Micrococcus luteus ATCC9341 was used to test urine at the time of slaughter for the presence of antimicrobial substances. Thirty-seven (7.4%) of the urine samples were positive for antimicrobial substances. Positive cases were not distributed according to breed (P>0.05), but occurred significantly more in male cattle aged 3-9 years and in those with good body condition score (P<0.05). Thirty-eight cases, in the form of 16 different pathological conditions, were seen from visual inspection of carcasses. Lymphadenopathy was seen in 10 (23.7%) carcasses including one antimicrobial substances positive carcass. Dermatophilosis and emaciation constituted 15.8 and 13.2% of pathological conditions observed, respectively.

Vascular endothelial growth factor (VEGF) stimulates the tyrosine phosphorylation of focal adhesion kinase (FAK), increases focal adhesion formation and is chemotactic for human umbilical-vein endothelial cells (HUVECs). In the present study we identified the major sites of VEGF-induced FAK tyrosine phosphorylation and investigated the mechanism mediating this pathway in the action of VEGF. VEGF increased the focal adhesion localization of FAK phosphorylated at Tyr-397 (Y397) and Y861 but stimulated a marked increase in phosphorylation at Y861 without significantly affecting the total level of phospho-Y397 FAK. Inhibition of Src with the specific inhibitor 4-amino-5-(4-chlorophenyl)-7-(t-butyl)pyrazolo[3,4-d]pyrimidine (PP2) completely blocked VEGF-induced Y861 phosphorylation without decreasing the level of phospho-Y397 FAK. We also examined the role of Src in mediating endothelial functions of VEGF in which FAK has been implicated as having a role. PP2 markedly inhibited VEGF-induced chemotaxis and wound-healing cell migration. The Src inhibitor also decreased the anti-apoptotic effect of VEGF determined by surface staining of annexin V but did not increase FAK proteolysis or prevent the VEGF-dependent inhibition of FAK proteolysis. In contrast, the specific PtdIns 3-kinase inhibitor LY294002 induced apoptosis and markedly decreased p125(FAK) expression and increased FAK proteolysis but had little effect on Y861 phosphorylation. These findings identify Src-dependent FAK phosphorylation at Y861 as a novel VEGF-induced signalling pathway in endothelial cells and suggest that this pathway might be involved in the mechanisms mediating VEGF-induced endothelial cell migration and anti-apoptosis.
